The Detective Conan series features characters created by Gosho Aoyama. In anticipation of Gin and Vodka investigating his whereabouts, Shinichi uses the pseudonym Conan Edogawa as his new identity. He then lives with his childhood friend, Ran Mouri and helps his father Kogoro - a private detective, to solve cases, in hopes of finding the case involved with the Black Organization. Only a few people know Conan's true identity. When he is disguised as Conan, he shows an innocent and cute character, like a 1st grader. His signature greeting is "Ah-re-re?!" (Lhoo?), an expression ala elementary school children when they see an oddity.
On the other hand, while investigating cases as a high school detective, he looks serious and has high intuition like an adult. In the English version of the comic - Case Closed, Shinichi Kudo's name is Jimmy Kudo. In the Japanese version of the animation, the voice of Shinichi is played by Kappei Yamaguchi, while the voice of Conan is played by Minami Takayama. In the live-action drama special, Shinichi Kudo is played by Shun Oguri, and later by Mizobata Junpei. Ran Mouri (毛利, Mōri Ran) is Shinichi Kudo's childhood friend and first love. Throughout the story, Ran always keeps her feelings for Shinichi and is faithfully waiting for his return. Ran lives with her father, Kogoro Mouri, who is a private detective. Although still in high school, Ran is reliable in household matters, and is good at taking care of her father who lives separately from his mother. One of Ran's close friends is Sonoko Suzuki, who is often a counselor in her love affairs. Ran is skilled in karate, but is afraid of ghosts. He is the chairman of the karate club and has an attractive stature. Although strong, Ran is also described as a character who cries easily. Ran several times almost revealed Conan Edogawa's real identity. In the English version, Ran is named Rachel Moore. In the Japanese version of the animation, the voice of Ran is played by Wakana Yamazaki. In the live-action drama special, Ran Mouri was played by Tomoka Kurokawa, but was replaced by Kutsuna Shiori. He sometimes visits Japan to take care of his son. Yukiko is Vermouth's best friend when she was an actress. Yukiko's voice in the Japanese animation is Sumi Shimamoto and her English voice is Laurie Steele. In Case Closed, Yukiko Kudo is known as Vivian Kudo. Eri Kisaki (妃, Kisaki Eri) is Ran Mouri's mother and Kogoro Mouri's wife. She worked as a lawyer and was known as the "Queen of Lawyers". Eri lives separately from Kogoro, who is now living with Ran. Despite living separately, Eri is still seen wearing her wedding ring. Several times in the story he is described as still loving Kogoro even though he hates his husband's bad habits.

Kid often appears in Detective Conan's stories, especially when Jirokichi Suzuki appears who likes to announce the challenge of stealing precious stones. Conan is always there to solve and thwart Kid theft tricks. In the story, Conan and Kid's relationship is described as rivals. Meanwhile in Detective Conan's story film, Kid is depicted knowing Conan's real identity. Starting from volume 95 of the Detective Conan comic, Kid is depicted knowing Conan is a high school detective. Kid the Thief is actually the protagonist in Aoyama Gosho's Magic Kaito series. His real name is Kaito Kuroba (黒羽, Kuroba Kaito). He has a face and stature that is very similar to Shinichi Kudo, so Ran once mistakenly recognized him as Shinichi. He is described as interested in Kogoro Mouri and has analytical skills like a police detective. Tooru works as a cafe waiter at Poirot's Café and studies under Kogoro. In volume 76 it was revealed that Tooru Amuro is a secret agent of the Tokyo Police Public Security Bureau named Rei Furuya (降谷, Furuya Rei). Tooru's voice in the Japanese animation is Tohru Furuya. Mary Sera (メアリー, Mearī Sera) is a mysterious character who first appears in volume 83. Mary has the stature of a middle schooler, but has strong intuition and is good at martial arts like an adult. Mary's character is depicted always hiding in a hotel room and only interacting with Masumi Sera, her son. In chapter 1049 it is finally revealed that he has blood relations with the Miyano family after Vermouth put the drug APTX 4869 into Mary's mouth and said "How does it feel to leave this world in the hands of your sister's creation".

Rumi Wakasa (若狭留美 Wakasa Rumi) is a character who is also introduced into the story, when Conan is investigating the whereabouts of a member of the Black Organization codenamed Rum. He works as an elementary teacher at Teitan Elementary School and has a clumsy, yet mysterious personality. Rumi is seen to experience frequent personality changes and has a high level of martial arts skills. He is described as having an interest in Shinichi Kudo and has a list of victims of the APTX4869 poison on his laptop.
